Chembio DPP® Syphilis Screen & Confirm – CE-marked dual rapid test

Chembio DPP® Syphilis Screen & Confirm represents a new standard in point-of-care diagnostics for syphilis. The test enables both screening and confirmation in the same cassette, as it separately measures treponemal (T) and non-treponemal (NT) antibodies. This provides a clearer indication of whether there is an active infection or a past syphilis infection and allows treatment decisions to be made quickly and close to the patient.

Function and method

The test is based on Chembio’s Dual Path Platform® (DPP®) lateral-flow technology. A small sample volume is applied to the sample well, followed by buffer. After approximately 15 minutes, two independent lines develop: the T line for treponemal antibodies and the NT line for non-treponemal antibodies. A control line confirms correct test performance.

Performance and accuracy

In large datasets with more than 1,600 serum samples, the T line showed high concordance with TP-PA (approx. 96.5% reactive and 95.5% non-reactive). The NT line had particularly high sensitivity at RPR titer levels of 1:8 or higher (almost 100%) and a specificity of around 98.6%. Independent studies have shown that use of the DPP® Micro Reader increases sensitivity, especially at low titers where visual interpretation may be less reliable.

In populations with many low titers, it is recommended to use the DPP® Micro Reader or supplement with laboratory testing, depending on local guidelines.
