BIOSYNEX CryptoPS is a rapid, semi-quantitative immunochromatographic test for the detection of capsular antigens from Cryptococcus species (CrAg) in capillary/venous whole blood, serum, plasma and cerebrospinal fluid (CSF). The test provides a visual result in approx. 10 minutes and is supplied with a positive control for reliable quality control. A simple titration procedure makes it possible to identify high antigen titers directly.
The readout is two-tiered: the T1 line appears at ≥25 ng/mL CrAg in CSF and at ≥50 ng/mL in serum, plasma and whole blood; the T2 line appears at ≥2500 ng/mL. The semi-quantitative readout helps the laboratory distinguish between low and high concentrations and can thus support the clinical assessment of patients in risk groups.
Cryptococcosis is an opportunistic fungal infection, most often caused by Cryptococcus neoformans, and is seen particularly in immunosuppressed patients. The most common clinical manifestation is meningoencephalitis. Rapid detection of CrAg can therefore be crucial in diagnosis and follow-up.
Performance and validation
In internal comparison with another commercial rapid test, a specificity of 100% and a sensitivity of 95% are reported (see instructions for use for details and limitations). The test was developed in collaboration with the French National Reference Center for Invasive Mycoses and Antifungals (Institut Pasteur) and evaluated by the Department of Clinical Microbiology and Infectious Diseases, University of the Witwatersrand (ECCMID, 2016).
How to use the test
Collect 20 µL sample (whole blood/serum/plasma/CSF), add three drops of dilution buffer and read the result visually after 10 minutes. A positive result may appear earlier. The product is an in vitro diagnostic device; read the instructions for use carefully before use.
